I know there are "question" words, but how do I turn a word into a question, like using an English question mark? For instance, how would I turn Qo'noS into a question. Like, if someone said, "We are approaching Qo'noS", and I wanted to be sure I had heard them right, how would I say, "Qo'noS?"?

In Star Trek III, Kruge responds to the news that the Federation rescue ship is the Enterprise with the exclamation 'entepray'...'a'?, subtitled Enterprise?. This usage is not described anywhere, but it seems a reasonable example to emulate if necessary.

I'll take your word for it. It's been a long time since I watched it. Anyway....I would say, "Qo'noS'a'?" or "Qo'noS...'a'?"

Oh, okay. I just found it. If you look in the back of TKD it tells you how to use 'a' properly.
